Why longevity matters

Living longer is only half the story.

What would you do with more years of strength, clarity and possibility? Longevity science asks how we might better understand the biology of aging and extend healthspan — the period of life spent in good health.

Our focus is the years in which people remain capable, engaged and independent. We do not promise a specific number of additional years to anyone. The honest goal is a longer stretch of life in which another first is still possible.

The Klotho story

Meet Klotho.

Klotho is a longevity-associated protein produced mainly in the kidneys and also expressed in the brain — named for the Greek goddess who spins the thread of life. It has roles in mineral metabolism and has been studied in connection with cellular stress, cardiovascular biology, cognition and aging. In plain terms: it helps regulate mineral balance, has been studied for its role in protecting cells from stress, and is an active research subject in brain and heart health. Klotho levels and expression decline with age, making the pathway an active area of longevity research.

The Klotho pathway

The Klotho Pathway, in four stages. One, aging: chronological aging naturally progresses. Two, declining Klotho levels: with age, Klotho levels and expression decline. Three, biological consequences: inflammation, oxidative stress, mitochondrial dysfunction, cellular senescence, tissue dysfunction and reduced resilience. Four, restoring Klotho signalling, an investigational strategy: restoring Klotho signalling may support healthy aging, resilience and longevity.

The Klothosome concept — investigational

The frontier is delivery.

Klotho is a large, fragile protein, and getting it where it needs to go is the hard part. Extracellular vesicles are tiny lipid-bound packages cells naturally use to ship proteins and genetic material to one another — a delivery system the body already recognises. Load Klotho into an extracellular vesicle and you have what the field has begun calling a Klothosome. The Klothosome concept explores whether an engineered extracellular-vesicle approach could serve as a potential delivery strategy for Klotho.
